Yes/no. The anime is based on a decades-old manga by the late father of Japanese comics/animation, Osamu Tezuka. But, his visual inspiration was the famous 'Metropolis' poster featuring the robot Maria. That's about all they have in common.
Is not. The cityscapes are totally 1931 metropolis-ish. Of course, by now those cityscapes are part of the collective unconscious, as evidenced by the 5th element. Clearly I will just have to watch the damn thing.
Well, he's definitely working with an art deco vision of the future, but I don't think that necessarily implies that he copied it from the film. (It's not like Fritz Lang invented what he was doing in 1927, either.) However, it's also possible that Otomo Katsuhiro ("Akira"), who created the film version, added more Lang-style elements. Regardless, it's a neat movie with great music.
